The Centrifuge Reagent Volume Calculator estimates how much reagent or resuspension solution is needed across a centrifugation batch when each tube, sample, or pellet receives the same volume. It multiplies the per-unit volume by the number of units and adds an optional extra percentage for transfer loss or preparation overhead.

Use it for buffer, wash solution, lysis solution, resuspension medium, or another protocol-defined liquid associated with a centrifuge step. The calculator does not determine centrifuge speed, relative centrifugal force, rotor compatibility, fill limits, or a scientifically appropriate reagent recipe; those parameters must come from the applicable method and equipment instructions.

1. Count the centrifuge units

2. Enter volume per unit
Use the protocol-defined amount added to one unit, in mL.

3. Add preparation overhead if needed
Set a percentage for expected transfer loss or vessel dead volume. Enter 0 for the exact theoretical requirement.

Prepare the reported total only after confirming the reagent formulation and per-unit volume in your protocol.

5. Keep centrifuge settings separate
Use the equipment method for rotor, balance, speed or RCF, time, temperature, and tube limits; this page calculates liquid volume only.

Base reagent volume = number of units × reagent volume per unit

Extra volume = base reagent volume × extra %

Total reagent = base reagent volume + extra volume

  • Number of units = tubes, samples, or pellets receiving reagent.
  • Reagent per unit = mL allocated to each unit.
  • Extra % = optional overhead for preparation and handling.

Assumptions: The same reagent volume is assumed for every counted unit. The calculation is independent of centrifuge speed, rotor radius, and run time.

What the result means

The result is the total volume to prepare for the entered number of centrifuge-related units, including the chosen overhead.

Check tube fill volumes, reagent compatibility, and protocol-specific handling before use.

Given:

  • 36 tubes
  • Reagent per tube = 0.75 mL
  • Extra allowance = 6%

Calculation:
Base = 36 × 0.75 = 27.0 mL
Extra = 27.0 × 0.06 = 1.62 mL
Total = 27.0 + 1.62 = 28.62 mL

Result:
28.62 mL

This is the preparation volume for the liquid step; centrifuge operating parameters are not part of the calculation.

Does this calculate RCF or RPM?

No. It calculates reagent volume only. Centrifuge force or speed requires rotor information and a different calculation.

What can the per-unit reagent represent?

It can represent a protocol-defined wash, resuspension, lysis, buffer, or other liquid volume applied equally to each counted unit.

Should I include spare tubes in the count?

Include only units you actually expect to dispense reagent into, unless you intentionally want extra complete aliquots. General overhead is better represented by the extra percentage.

Can I enter microliters?

This page uses mL. Convert microliters to mL by dividing by 1,000 before entering the per-unit volume.

Does extra reagent change the amount added to each tube?

No. The per-unit amount remains the same. The extra percentage increases the batch prepared so handling losses are less likely to leave the final units short.
