Bacterial Growth Sample Capacity Estimator

The Bacterial Growth Sample Capacity Estimator calculates how many complete samples can be prepared from an available bacterial culture volume when each sample requires a fixed aliquot. It can also reserve a specified percentage for pipetting loss, dead volume, repeats, or other planned overhead.

This is useful when allocating cultures across assays, replicate wells, tubes, or downstream preparations. The calculator reports the usable volume after reserve, the number of whole samples that fit, and the leftover usable volume. It does not model cell concentration requirements; if each sample must contain a target number of cells, first determine the required aliquot volume from concentration.

Reserved volume = available volume × reserve %

Usable volume = available volume − reserved volume

Sample capacity = floor(usable volume / volume per sample)

Leftover = usable volume − sample capacity × volume per sample

Where:

  • Available volume and volume per sample are in mL.
  • Reserve % is entered as a percent and converted to a decimal.
  • floor means round down to the nearest whole sample.

Assumptions: All samples are assumed to use the same culture volume and the reserve is taken from the available volume before samples are counted.

What the result means

The main result is the maximum number of complete equal-volume samples that fit after the reserve allowance.

This is a volume capacity estimate; confirm concentration, sterility, and protocol-specific minimum volumes separately.

Given:

  • Available culture = 84 mL
  • Volume per sample = 2.5 mL
  • Reserve = 8%

Calculation:
Reserved = 84 × 0.08 = 6.72 mL
Usable = 84 − 6.72 = 77.28 mL
Capacity = floor(77.28 / 2.5) = 30 samples
Leftover = 77.28 − 30 × 2.5 = 2.28 mL

Result:
30 complete samples, with 2.28 mL usable volume left

The reserve is protected first, then only fully supplied samples are counted.

Why does the calculator round the sample count down?

A fraction of the required aliquot does not make a complete sample. Rounding down avoids overstating how many fully supplied samples can be prepared.

What should I include in the reserve percentage?

Use it for expected pipetting losses, vessel dead volume, QC aliquots, or planned repeats as appropriate to your workflow. The calculator does not prescribe a universal reserve.

Can I enter microliters?

This page uses mL. Convert both the available volume and per-sample volume to mL so the units are consistent.

Does this account for target cells per sample?

No. It allocates volume only. If samples require a minimum cell number, determine the necessary volume from culture concentration and use that as the per-sample input.

What does leftover volume exclude?

Leftover is calculated from the usable portion after the reserve has already been removed. The reserved volume is shown separately in the breakdown.
