The DNA Concentration Reagent Volume Calculator determines how much DNA stock is needed to deliver a specified DNA mass per reaction across a chosen number of reactions. It is designed for routine setup work where concentration is known in ng/µL and the protocol calls for a target quantity of DNA.

Because the required stock volume is simply target mass divided by stock concentration, the calculator is useful for quickly checking whether a planned pipetting volume is practical. The page reports both the volume required for one reaction and the total volume for all reactions, along with the total DNA mass represented by the batch. It does not automatically add overage for pipetting loss, so any desired excess should be reflected in the reaction count or target plan.

The mass and concentration units must be compatible. With ng and ng/µL, the calculated volume is in µL.

What the result means

The main result is the volume of DNA stock needed to supply the target DNA mass to every entered reaction.

If your protocol requires extra volume for transfer loss or a master mix, add that overage separately.

Given: DNA stock concentration is 32 ng/µL, each reaction needs 80 ng, and 18 reactions are planned.

Calculation:
Volume per reaction = 80 ÷ 32 = 2.5 µL.
Total volume = 2.5 × 18 = 45 µL.
Total DNA mass = 80 × 18 = 1,440 ng.

Result: 45 µL of DNA stock is required.

Each reaction receives 2.5 µL of the 32 ng/µL stock to supply 80 ng of DNA.

What if my concentration is in µg/mL?

Numerically, 1 µg/mL equals 1 ng/µL, so the same numeric concentration can be used when the units are converted consistently.

Can the target mass be zero?

Yes, but the calculated DNA volume will also be zero. For a real DNA-containing reaction, enter the positive mass required by the protocol.

Why is the required volume very small?

A high-concentration stock can produce sub-microliter volumes. If that is impractical to pipette accurately, prepare a working dilution before setup.

Does the calculator include pipetting overage?

No. It multiplies the exact per-reaction requirement by the reaction count; add extra reactions or prepare an overage separately if needed.

Is total reaction volume part of this calculation?

No. This tool calculates only the DNA stock volume needed to meet a mass target, not the complete reaction mixture volume.
